For estimation of growth, the efficiency of linked cross sectional scheme has been compared with pure longitudinal and cross sectional schemes. The relevant estimation theory has been developed and the expressions for the optimum estimators alongwith their variances have been derived. It has been observed that for estimation of growth, the linked cross-sectional scheme has been observed to be less efficient as compared to pure longitudinal scheme but more efficient than pure cross-sectional scheme.  相似文献

The effect of mixing on biogas production of a 1.5‐m3 pilot continuous stirred tank reactor (CSTR) processing screened dairy manure was evaluated. Mixing was carried out by recirculation of reactor content with a mono pump. The experiment was conducted at a controlled temperature of 37±1°C and hydraulic retention times (HRTs) of 20 and 10 days. The effect of continuous and intermittent operation of the recirculation pump on biogas production was studied. At 10 days of HRT, the results showed a minimal influence of recirculation rate on biogas production and that continuous recirculation did not improve reactor performance. At 20 days of HRT, the recirculation rate did not affect reactor performance. Combination of low solid content in feed animal slurry and long HRTs results in minimal mixing requirements for anaerobic digestion.  相似文献

Accurate prediction of the leaching requirements (Lr) of crops and striving to attain them is essential for efficient irrigation water use. Solute modeling was extended to develop four Lr conceptual models that do not neglect solute reactions in the root-zone, surface evaporation, and the influence of immobile wetted pore space. The models were based on: (i) the water movement equation which included an exponential water-uptake function (-e) or the 40-30-20-10 water-uptake function (-4); (ii) the solute movement equation for a reactive salt of a linear reaction term (the Lrchem-e and Lrchem-4 models); or the employment of output (salinity of soil solution, EC vs concentration factor, CF) of the SAO comprehensive chemical model (the LrSAO-e and LrSAO-4 models); and (iii) the inclusion of an effective soil solution volume in the transport equations. The root-zone average relative effective soil solution volume νeff (L | L50, p) was of sigmoidal response to leaching fraction (L) with two adjustable parameters L50 and p; the root-zone average reduced retention coefficient decreased linearly with L; and salt concentration at soil surface was related to salt concentration of irrigation water (ECi) by the fraction of irrigation water that evaporated (∈). The resulted concentration profiles indicated the salt behaved as a conservative one down to a threshold depth (xs) below of which salt was retained and precipitated. The depth of the conservative-salt front, xs increased with L and the 40-30-20-10 water-uptake pattern overestimated the xs depth relative to the exponential pattern. Concentration profiles were integrated to compute the root-zone average salinity, which was converted to crop salt-tolerance threshold (AE). The four conceptual models were successfully calibrated using experimental AE/ECi vs. Lr data with the input parameter values: ς = 0.27, p = 1.44, L50 = 0.16, ω = 2, and ∈ = 0 or 0.1 for the exponential or the 40-30-20-10 pattern, respectively; where ς is relative root length parameter and ω is a weighing parameter. No significant difference existed between the four model correlations at the 0.05 level. The four models require ECi and AE of the crop as input for Lr prediction. Sensitivity analysis revealed predicted Lr was sensitive the least to error in ∈. For tolerant and moderately tolerant crops Lr was sensitive the most to ς, and for sensitive crops to L50 and p. Model verification and validation were discussed. In deriving the present Lr models, no osmotic adjustment was required and both the exponential and the 40-30-20-10 water uptake patterns were, equivalently, applicable. Abstract A cellulase-containing fraction present in the culture fluid of Trichoderma reesei grown on cellulose was obtained by fractionated centrifugation. The buoyant density of this fraction was D = 1.060 g/ml. Its ultrastructural properties, as detected by transmission electron microscopy, are given. The fraction consists of membrane vesicles attached to a carbohydrate polymer. This polymer is positive to Ruthenium red staining.
The effect of urea on the extraction and separation of acidic cellulases from this fraction is described. Linear gradient gels for both urea (up to 8.0 M urea) and polyacrylamide gels (up to 30%) were used to determine adequate separation conditions for isoelectric focusing (IEF) in a polyacrylamide gel matrix. The effect of urea on the extraction and separation conditions was tested by titration curves. In the presence of 6.0−8.0 M urea, the main cellulase-containing hydrolase complex (pIapp4.2) from this fraction is split into 3 isoenzymes and a further cellulase (pI 5.65).  相似文献

Summary The effect of cropping systems of wheat-maize (WM), wheat-rice (WR), wheat-groundnut (WG), gram-bajra (GrB), potato-guara (PGu), and raya-mash (RaMa) in combination with treatments of dummy (uncultivated area) and applied Zn 0.0 (Zn0), 2.8 (Zn1), 5.6 (Zn2) 11.2 (Zn3) kg/ha was studied on the transformation of labile Zn fractions: exchangeable (Exch.), adsorbed (TAd) [weakly (WAd), moderately (MAd), strongly (SAd)], and organic matter (OM) in different layers of sandy loam soil. The added Zn stayed largely in the 0–30 cm layer and was associated with the WAd- and OM-Zn fractions. About 70% of the total labile Zn (PAv) remained in the WAd- and OM-Zn, that is, 33 and 39% in 0–15 cm layer, and 33–39% and 31–36% in 16–150 cm layer. All the Zn fractions in 0–15 cm layer, and only of WAd in 16–30 cm layer, significantly increased with rates of Zn addition. These were also significantly higher in Zn1–3 than Zn0 and dummy treatments because of the residual Zn. Diverse effects of cropping systems on soil properties, residual Zn, and labile Zn fractions were found. The influence was strong in 0–15 cm layer decreasing gradually with soil depth due largely to differences in Zn requirement, crop intake of various Zn fractions and the cultural practices of the systems. All the crops and rotations appreciabilly responded to Zn application. Uptake of Zn by crops markedly and successively increased with increasing rates of Zn application. The WR caused a significant increase in soil organic matter whereas WR and WM in CaCO3. The WR, WM and GrB resulted in a decrease in pH while WG and GrB in CaCO3. The RaMa and PGu maintained much higher residual Zn than other systems. The systems which caused the maximum decrease in Zn fractions were: cereal-cereal (WM) in Exch. legume-millet (GrB) in all the adsorbed, PAv and the Zn associated with CaCO3, vegetable-legume (PGu) also in MAd and SAd; and cereal-legume (WG) in OM and PAv. Hence GrB, WG and WM in that order will cause the deficiency of Zn much earlier than the other systems due to greater use and or transformation of WAd- andOM-Zn. Such effects were least under RaMa because it increased the WAd-, MAd- and OM-Zn.  相似文献

Efficiency of linked cross-sectional scheme has been compared with pure longitudinal schemes for generalised estimation of norm relaxing the condition of constancy of variances. The relevant theory of estimation has been developed and the expression for the optimum estimators alongwith their variances have been derived. The percentage gain in efficiency has been calculated. It has been observed that for higher values of Q, percentage gain in efficiency of proposed scheme over cross-sectional is very high.  相似文献

This work presents a rapid and simple freeze centrifugation method to concentrate dilute protein solutions for detection by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS–PAGE) Coomassie blue staining. Moreover, a simple way to assemble a cryoconcentration device is presented, and its use is discussed. Commercial purified protein standard and an enzyme with high fructosyltransferase (FTase) activity, coming from target fractions obtained by chromatographic separation, were used as an example. FTase, coming directly from the chromatographic fractions, was difficult to view through SDS–PAGE analysis; however, it was easily visualized, and its activity was enhanced, after the application of the freeze centrifugation protocol presented here.  相似文献

Mechanisms of cellular reactions responsible for the spreading non-transformed cultured tissue cells on the surface of various substrata and relationships of these reactions to the control of cell proliferation are reviewed; the special role of the membrane-cystoskeleton interactions leading to extension and attachment of psuedopods is stressed.Transition of cells from non-transformed to transformed phenotype is characterized by decreased spreading and by decreased dependence of proliferation on spreading. Manifestations of both of these spreading-associated changes are reviewed and their possible mechanisms are discussed. It is suggested that cell transition to transformed phenotype involves shift of an equilibrium between the reactions induced by the two groups of membrane-bound ligands: those attached and those not attached to the substratum.  相似文献

Vegetation is a major environmental factor influencing habitat selection in bird species. High resolution mapping of vegetation cover is essential to model the distribution of populations and improve the management of breeding habitats. However, the task is challenging for grassland birds because microhabitat variations relevant at the territory scale cannot be measured continuously over large areas to delineate areas of higher suitability. Remote sensing may help to circumvent this problem. We addressed this issue by using SPOT 5 imagery and phytosociological data. We mapped grassland vegetation in a floodplain using two methods. We (i) mapped the continuous Ellenberg index of moisture and (ii) identified 5 vegetation classes distributed across the wetness gradient. These two methods produced consistent output maps, but they also provided complementary results. Ellenberg index is a valuable proxy for soil moisture while the class approach provided more information about vegetation structure, and possibly trophic resources. In spite of the apparent uniformity of meadows, our data show that birds do not settle randomly along the moisture and vegetation gradients. Overall birds tend to avoid the driest vegetation classes, i.e. the highest grounds. Thus, vegetation maps based on remote sensing could be valuable tools to study habitat selection and niche partition in grassland bird communities. It is also a valuable tool for conservation and habitat management.  相似文献
